Multidrug resistance reverting activity and antitumor profile of new phenothiazine derivatives

Abstract A series of easily affordable phenothiazine derivatives bearing a rigid but-2-ynyl amino side chain were synthesized and tested to evaluate the MDR reverting activity and full antitumor profile. Some compounds endowed with remarkable MDR reverting effect were identified, and the most active one ( 6c ) was shown to increase doxorubicin retention in multidrug resistant cells, suggesting a direct interaction with P-glycoprotein. Furthermore, a broad range of cellular activities were observed for different compounds. Continuing our studies on combilexines, compounds consisting of a DNA intercalator linked to a minor groove ligand, new results are presented. The synthesis of a series of new propylamine oligopyrrole carboxamides closely related to netropsin and distamycin A, linked to a heterocyclic or anthraquinone system is reported. The cytotoxic activity in vitro, the DNA binding characteristics and the inhibition of the topoisomerase I of the compounds were studied in order to explain the biological mechanism of action of these new potential combilexines. Modulation of intracellular calcium concentrations and T cell activation by prickly pear polyphenols.

Opuntia ficus indica (prickly pear) polyphenolic compounds (OFPC) triggered an increase in [Ca2+]i in human Jurkat T-cell lines. Furthermore, OFPC-induced rise in [Ca2+]i was significantly curtailed in calcium-free buffer (0% Ca2+) as compared to that in 100% Ca2+ medium. Preincubation of cells with tyrphostin A9, an inhibitor of Ca2+ release-activated Ca2+ (CRAC) channels, significantly diminished the OFPC-induced sustained response on the increases in [Ca2+]i. Lanthanum and nifedipine, the respective inhibitors of voltage-dependent and L-type calcium channels, failed to curtail significantly the OFPC-induced calcium response. La regione dei Caraibi abbraccia, infatti, sia alcune delle isole dell’arcipelago delle Piccole Antille (Martinica, Guadalupa), sia l’isola di Haïti, che fa parte delle Grandi Antille, che la Guiana incastonata sulla costa orientale dell’America meridionale. Pur nella diversità dei movimenti ideologici e degli avvenimenti storici che si sono succeduti nella regione, le opere letterarie degli scrittori caraibici francofoni, nel loro insieme, hanno oscillato, e continuano …

The definition "Franco-Caribbean Literature" used by convention from the critique to indicate all the literary production in French of the central region of the American continent is very improper. The Caribbean region includes in fact the islands of Martinique Guadeloupe Haïti and the Guiana. Despite the diversity of ideological movements and historical events that took place in these countries the literary works of the Caribbean writers have fluctuated and continue to oscillate between two tendencies: that of rooting in tradition and that of escape from it. The writers show themselves open to comparison and dialogue with other literary and cultural traditions in order to create a narrative form that corresponds to the many inheritances of those composite populations. Here the cultural syncretism that has been created allows to recognize a common identity in the name of all that the West wanted to extinguish. The common denominator seems to be inverted in the contradictory result of courageous but dangerous encounters of curiosities that can lead to conflicts to exclusions.

This paper focuses on the story of two dark-skinned Italians written for Giorgio Marincola by Carlo Costa and Lorenzo Teodonio in Razza partigiana and for his sister Isabella by Wu Ming 2 and Antar Mohamed in Timira. Romanzo meticcio. The events of their lives as well as many other aspects of our colonial neocolonial and postcolonial history have re-emerged from the darkness in which they had been hidden only in the last fifteen years thanks to the reopening of the archives allowing to re-discuss many of the aspects that in the pages of History as well as in those of literature had been told in another way. Nonsteroidal anti-inflammatory drugs (NSAID) inhibit cyclooxygenase activity and cause gastrointestinal damage in part by promoting leukocyte accumulation in the mucosa. Our aim was to evaluate the effects of selective blockade of the isoenzymes cyclooxygenase-1 and cyclooxygenase-2 on leukocyte adhesion in vivo. Sensorimotor plasticity is defined as the ability to produce an appropriate motor response face to environmental or bodily changes. The classical experimental paradigm to study sensorimotor plasticity is prism adaptation, which consists of pointing to visual targets while wearing prisms that shift the visual field laterally or vertically. Effects of hydrazyl group containing drugs on leucocyte functions: an immunoregulatory model for the hydralazine-induced lupus-like syndrome.

Isoniazid (INH) and hydralazine (HYD) are transglutaminase (TGase, E.C.2.3.2.13.) substrates containing catalytically recruitable hydrazyl groups. Since they can be expected to inhibit TGase-mediated cell functions by competing with physiological substrates, their effect upon allogeneically and lectin-induced proliferation of mononucleocytes and upon zymosan-induced chemiluminescence of phagocytes was studied. Both compounds inhibited chemiluminescence in a dose-dependent manner. ID50 of HYD was consistently below 20 microM, while that of INH was above 120 microM.
